Maya SpeakTiki is a tiki-themed speakeasy hidden behind a tequila shelf inside Agave Bandido, a modern Mexican restaurant in the Hamlin area of Winter Garden. The vibes are fantastic once you are through the secret entrance, and the restaurant out front is worth a visit on its own: the food at Agave Bandido is genuinely great, so it is easy to turn this into a full dinner-and-drinks night.

Noir, Hidden Inside Talay
Noir is the classiest of the three: an intimate, low-lit cocktail room hidden behind an unmarked door through Talay, a Thai restaurant in downtown Orlando. The cocktails lean Thai-inspired, and unlike a lot of speakeasies, Noir serves its own menu of Thai-style tapas, so you can actually settle in and make a night of it rather than just stopping for one drink.
